Description
This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Twenty-four healthy volunteers of all races, twelve females and twelve males, will help determine how age, gender and hormonal status influence drug absorption in the body. The Researchers hope to prove that women handle medicines differently than men and that older women handle medicines differently than younger women. This may affect how medications are prescribed for women. To determine if they are eligible for the study, subjects will complete a questionnaire, undergo a physical examination and have blood drawn. They will be asked to volunteer for genetic testing for which additional blood will be drawn. This genetic testing looks at genes responsible for drug absorption. If they qualify, subjects will be admitted to the General Clinical Research Center (GCRC). Half will start with phase I and half will start with phase II. After 28 days subjects from phase I will go to phase II and subjects from phase II will go to phase I. Phase I will evaluate how digoxin given through an IV tube is absorbed. Phase II will evaluate how digoxin taken by mouth is absorbed. Phase I: Subjects will receive digoxin through a catheter tube in their arm. Blood will be drawn and urine specimens will be taken over the next three days. Only during phase I will Subjects have a procedure called an Esophagogastroduodenoscopy (EGD) to take tissue samples from the small intestine. Phase II: Subjects will receive a pill containing digoxin. Blood will be drawn and urine specimens will be taken over the next three days.
