This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. No clinical imaging method currently provides sufficient specificity to reliably differentiate between benign and malignant breast lumps. Pathologic evaluation is the only way currently to obtain a definitive diagnosis. The general goal of this research is to develop and test improved high field (4 Tesla) MRS methods, and to combine them with contrast-enhanced (Gd-DTPA) MRI, in studies of breast cancer. While MRS holds great promise for profiling the metabolic status of cancer cells, it has not yet been determined whether MRS detection of certain metabolites in vivo can provide high diagnostic specificity for differentiating between benign and malignant lesions. We believe that the ability to identify malignant lesions can be significantly improved using in vivo MRS. By implementing new methods that exploit the high magnetic field, optimize sensitivity, and separate certain metabolite resonances (choline compounds and 13C labeled lactate) from overlapping macromolecular resonances (lipids), false-positive and false-negative diagnoses can be avoided.
