Collagenesis and elastogenesis in fibrin-based media-equivalents treated with TGF-ßl and Insulin

Abstract

The neonatal smooth muscle cells (SMC) produced more collagen when seeded in fibrin gel rather than collagen gel. The quantity of cross-linked elastic fibers was enhanced by treatment with TGFβ1 and insulin. The When compared to collagen ME , the fibrin ME compacted to a greater extent and were both stronger and stiffer when cultured under the same conditions, having a tensile modulus and ultimate tensile strength (UTS). It was found that the use of soluble factors increased elastin production, as measured by alkaline extraction and amino acid analysis, 5-fold in fibrin gels, which was about 4 times greater than in collagen gels.
