COMBINED ON-CHIP SPECTROSCOPY AND RHEOLOGY AS A PRECLINICAL DRUG SCREENING TOOL FOR SICKLE CELL DISEASE

Abstract

Combined studies in blood rheological behavior and hemoglobin oxygen saturation can help in understanding investigational drugs for sickle cell disease (SCD) from both a therapeutic and mechanistic perspective. We have modified a microfluidic platform to evaluate rheological behavior and changes in hemoglobin (Hb) oxygen affinity after treatment with oxygen affinity-shifting compounds. The results show both a reduction in the oxygen-dependent rheology of SCD blood and increased Hb oxygen affinity. This platform improves upon previous devices that solely analyze the fluid behavior of blood in that it can identify beneficial shifts in Hb oxygen affinity without the need for additional testing.
