Development of a multiphoton fluorescence lifetime imaging microscopy system using a streak camera

Abstract

The development and calibration of a multiphoton fluorescence lifetime imaging microscopy (FLIM) system was described. FLIM studies were carried out on cellular specimens such as stained pollen grains and fibroblast cells. The lifetime values were found to correspond well with results of earlier studies on the same specimens. Measurement of intracellular physiology and fluorescence resonance energy transfer imaging were identified as potential applications for the system.
